Amanda Serrano will return to the ring on May 30 at the El Paso County Coliseum in Texas.
She will defend her WBA and WBO titles against Germany's Cheyenne Hanson on the same bill as Stephanie Han's rematch with Holly Holm for the WBA lightweight title.
Also on the bill, Desley Robinson puts the IBF and WBO middleweight titles on the line against Mary Spencer while Yokasta Valle moves back to light-flyweight to box Lourdes Juarez for the WBC title.
"I respect every opponent who steps in the ring, and I know Hanson is coming to win and she has KO power," Serrano said.
"I'm also continuing to hunt the all-time knockout record, so that's always on my mind, but everything starts with discipline, execution, and performing at the highest level on fight night.
"Stephanie Han is all class, and it is my pleasure to come to her city and make the walk before her on Saturday May 30. See you soon El Paso."
Serrano is the first-ever Puerto Rican undisputed world champion and the most decorated fighter in women's boxing history.
She fought Katie Taylor in one of the sport's most high-profile trilogies.
Serrano made a long-awaited homecoming to Puerto Rico in a unified featherweight title defence in January, defeating challenger Reina Tellez.
